A Valentine, Arthur trova i gemelli Proetus e Acrisius mentre litigano per Helen. Ciascuno sostiene di essere l'uomo più coraggioso e devoto, mentre Helen ha poca pazienza per entrambe le pretese. I fratelli chiedono ad Arthur di decidere la loro prima sfida. A turno si mettono in mostra con le pistole, e Arthur fa da testimone alla loro rivalità. Nessuno dei due gemelli accetta che l'altro abbia dimostrato qualcosa, quindi la sfida resta irrisolta quando si separano.
Arthur incontra di nuovo Proetus e Acrisius più tardi, mentre preparano un'esibizione più pericolosa per Helen. La loro gara di tiro lascia il posto a una prova di coraggio con barili e acqua impetuosa. Un fratello parte per primo, poi l'altro lo segue, deciso a non essere da meno. Arthur osserva il loro folle tentativo e aiuta i fratelli a superarlo sani e salvi. Helen si rifiuta ancora di scegliere tra loro, e la competizione continua.
Per la loro ultima sfida, Proetus e Acrisius organizzano un esperimento elettrico e chiedono ad Arthur di azionarlo. Ciascun gemello si siede nell'apparato, convinto di poter sopportare più dolore del fratello. Arthur attiva la macchina mentre Helen osserva la loro esibizione. La sua risposta delude entrambi. - ◆In "Oh, Brother I", if the player kills one of the twins, the other will say their name in shock as he and Helen look on in horror. The mission will then fail.
- ◆One of the brothers hands Arthur cash at the end of "Oh, Brother I", but no amount is added to his inventory.
- ◆Unlike most stranger missions, when the first (or first and second) encounter is completed by Arthur and the arc is finished by John, there is no unique dialogue addressing John being a friend of Arthur's who passed. The twins will act as if they were already acquainted with John beforehand.
- ◆There appears to be a bug where, during "Oh, Brother II", Proetus' and/or Acrisius' dialogue will emit from the wrong brother, while the correct brother's mouth moves.
- ◆If the player kills the twins before entering "Oh, Brother I", bystanders and lawmen will ignore the crime.
- ◆If "Oh, Brother I" is entered without any sidearm, the protagonist will freeze, leaving abandonment of the mission as the only option.
- ◆Mission "Oh, Brother II" can be failed, if one of the brothers die. However, performing that is near impossible, as the player is locked out of their weapons.
- ◆In the game files there is a removed ped named CS_twingroupie_02, a unused female character that was part of the mission.
- ◆In Greek mythology, Acrisius and Proetus were brothers who quarreled even in their mother's womb. When their father (Abas) died, Acrisius expelled Proetus from his inheritance and became king of Argos. Proetus later returned, and Acrisius was compelled to share his kingdom with his brother by giving Tiryns to him, while he retained Argos for himself. Acrisius was the grandfather of Perseus, who would later kill him.
- ◆If the player uses trainers on PC to kill Helen in "Oh Brother I", the twins will exclaim her name as she dies, and a hidden animation triggers where Proetus tries to attack the player but Acrisius holds him back, and the mission fails shortly after.
