Nach Friends Reunited öffnet Flight School am Exceptionalists Way beim Los Santos International Airport. Michael De Santa oder Franklin Clinton kommt zur Schule und erhält von Jackson Unterricht, um seine Flugfertigkeit zu verbessern. Trevor Philips ist bereits ein erfahrener Pilot, kann den Kurs aber ebenfalls besuchen. Der Kurs beginnt in einer Cuban 800 und lehrt zunächst einen Start durch die markierten Tore, gefolgt von einer Landung auf der Landebahn. Anschließend geht es mit der Mallard weiter, um Rückenflug, Messerflug und eine Route in geringer Höhe über den Port of Los Santos und LSIA zu üben.
Die Lektionen mit der Mallard werden am Lago Zancudo fortgesetzt, wo der Pilot das Flugzeug auf einer Brücke landet und anschließend einen Innenlooping fliegt. Flight School wechselt dann zu einer Frogger für einen Hubschrauberkurs über Los Santos und eine schnellere Route vom Palomino Highlands zurück in Richtung Stadt. Danach schickt Jackson den Piloten zum Fallschirmspringen auf ein festes Ziel, gefolgt von einem Sprung auf ein bewegliches Ziel. Dom Beasley erscheint in den Einführungen zu beiden Fallschirmübungen.
Die letzte Lektion kehrt für Earn Your Wings zur Mallard zurück. Michael, Franklin oder Trevor fliegt einen Kurs durch Los Santos und nutzt an den grünen Toren Messerflug sowie an den blauen Toren Rückenflug, wodurch die während der Schule gelehrten Manöver kombiniert werden. Die zwölf Lektionen enden, sobald das Flugzeug diesen Stadtkurs absolviert hat. Nachdem jede Lektion abgeschlossen wurde, erscheinen Dusters am westlichen Ende des Marina Drive in der Grand Senora Desert und neben dem Alamo Sea.

- ◆The Flight School's logo, being an image of two hands clasped as if for prayer, backed by a pair of wings, is a visual pun on the phrase "on a wing and a prayer", used to describe the trust passengers of aircraft have towards the plane or helicopter's crew to safely respond to a developing incident.
- ◆Dom Beasley appears in the video introduction of the Skydiving and Drop Zone lessons.
- ◆Although Trevor is a trained pilot due to his military experience, he can still go to the Flight School and complete the lessons to reach 100% flying skill faster.

- ◆In the demonstration video of Engine Failure, the instructor claims that this course had lost a lot of pilots, which is an obvious reference to the large presence of Fire Trucks and Ambulances just in case of an accident.
- ◆In Moving Landing, the image depicts a Swift helicopter landing on the bed of a Flatbed. In the actual mission, the player has to land a Buzzard on the back of an Army Trailer.
