Der Brute Prison Bus ist ein Bus der Emergency-Klasse auf Basis eines Schulbustyps, der für den Gefangenentransport ausgelegt ist. Er bietet Platz für vier Personen. Seine lange, abgerundete Karosserie kombiniert eine Frontpartie für schwere Nutzfahrzeuge mit einem Busaufbau, der lose vom CV200 abgeleitet ist. Schwarze Verkleidungen bedecken Stoßfänger, Kühlergrill, Scheinwerfergehäuse und Spiegel, während vergitterte Fahrgastfenster, Warnleuchten auf dem Dach und eine hintere Notausgangstür ihm das Erscheinungsbild eines Justizvollzugsfahrzeugs verleihen.
In Grand Theft Auto V trägt der Prison Bus blaue Lackierung mit Emblemen des Bolingbroke Penitentiary State Prison, BPSP-Markierungen und einem weißen Karostreifen entlang der Dachseiten. Neben den vorderen Kotflügeln befinden sich Gruppe 6-Logos. Die Fahrerkabine besitzt in der Enhanced-Version analoge Instrumente im Lkw-Stil, und der Bus behält die Industriefelgen, die er mit mehreren kommerziellen Brute-Fahrzeugen teilt. Er ist für $731,500 erhältlich.
Der Prison Bus ist langsam und schwer, doch seine Masse und Panzerung erlauben ihm, Beschuss einzustecken und leichtere Fahrzeuge aus dem Weg zu schieben. Seine Höchstgeschwindigkeit ist für einen Bus hoch, allerdings sind die Bremsen schwach und das Handling durchschnittlich. In schnellen Kurven kann er auf die Seite kippen. Der Hinterradantrieb schränkt ihn an steilen Steigungen und im Gelände ein, doch seine Federung kommt weiterhin mit unebenem Untergrund zurecht. Das Spiel nutzt ihn als geschütztes Transportfahrzeug für Gefangene.

- ◆The Prison Bus, along with the Annihilator and Lifeguard, are the only police vehicles with a radio in single player.
- ◆The Prison Bus only seats four occupants like the normal Bus, but in the mission Off Route, Johnny and the six prisoners will be in the same vehicle.

This also causes an unintended effect: the animation for the third "service" will be distorted, as the prostitute will slowly move towards the driver's seat while performing the animation, then moving back to the seat when the service is done.

- ◆In the original version (PlayStation 3/Xbox 360) of the game, the bus was called "Police Prison Bus" in the HUD. In the enhanced version (PlayStation 4/Xbox One/PC) of the game, the bus is named simply "Prison Bus". The original name remains on the Rockstar Games Social Club.
