John Marston trifft Abraham Reyes in Agave Viejo, wo Reyes ihn bittet, ihm bei der Übernahme eines Versorgungszugs der Mexikanischen Armee zu helfen. Sie reiten durch den Regen nach Chuparosa. Reyes erklärt, dass der hintere Teil abgekoppelt werden muss, bevor der Zug abfährt, damit ihnen die Lokomotive und ihr Panzerwagen bleiben. Er sorgt für Ablenkung, während John sich unbemerkt auf den Bahnsteig schleicht, ohne Alarm auszulösen.
John beseitigt lautlos die Wachen entlang des Zugs, erreicht die Kupplung und trennt die hinteren Waggons ab. Während sie davonrollen, kümmert er sich um die Soldaten, die den verbliebenen Zug bewachen, darunter auch den Schützen an seinem Gatling gun. John räumt die Waggons vor sich frei, löst die Bremse der Lokomotive und übernimmt die Kontrolle. Reyes holt ihn ein, während der Zug sie aus Chuparosa fortbringt.
Während der Zug fährt, sammelt John Sprengstoff ein und klettert auf den Panzerwagen. Er bringt eine Ladung an dessen Luke an, zieht sich zurück und steigt nach der Explosion ein, die die Luke aufsprengt. Drinnen knackt er den Safe und nimmt dessen Inhalt an sich. John kehrt zu Reyes zurück, der sagt, er werde bald die Informationen beschaffen, die John braucht, und ihn dann neben dem Zug außerhalb von Casa Madrugada zurücklässt.

- ◆The name of this mission is inspired by the 1903 silent film The Great Train Robbery.
- ◆The plot of robbing a train loaded with ammunition is perhaps an allusion to Sam Peckinpah's film The Wild Bunch, which Rockstar noted as one of the films that inspired the game. In the film, however, the characters rob a train for a character not very unlike Colonel Allende.
- ◆This is the only time that a train is seen pulling more than five cars.
- ◆It always rains during this mission, possibly because the darkness of the clouds can allow John to approach more surreptitiously, and that the thunderstorm can help hide unnecessary noises.
- ◆This is the only Reyes mission which does not involve killing an antagonist. It is also the only Reyes mission that doesn't reward the player an Achievement/Trophy.
- ◆This is one of the few missions to require the player to neutralize an enemy without alerting anyone to their presence, another being At Home With Dutch.

- ◆After the train is uncoupled at Chuparosa, it rolls westwards, down the hill. The uncoupled train never actually stops, and continues to gain speed indefinitely. Using mods, it's possible to watch the unmanned train cross into the USA, making a loop around Gaptooth Breach and Tumbleweed, passing through Benedict Point, then re-entering Mexico. It will even pass its point of origin in Chuparosa, clipping through the stationary train, and zooming off through Mexico, back into the USA. By the time it has passed MacFarlane's Ranch and entered the tunnel in the Redemption Mountains, it is too fast to keep up with.
