John Marston se reúne con Abraham Reyes en Agave Viejo, donde Reyes le pide ayuda para apoderarse de un tren de suministros del Ejército Mexicano. Ambos cabalgan bajo la lluvia hasta Chuparosa. Reyes explica que deben desacoplar los vagones traseros antes de que el tren salga, dejando la locomotora y el vagón blindado para ellos. Él organiza una distracción mientras John se introduce en el andén sin poner en alerta a los soldados.
John acaba silenciosamente con los guardias que recorren el tren, llega al enganche y separa los últimos vagones. Mientras estos se alejan por la vía, se ocupa de los soldados que vigilan el resto del convoy, incluido el artillero de la Gatling gun. Tras despejar los vagones de delante, libera el freno de la locomotora y toma el control. Reyes alcanza el tren cuando ya se aleja de Chuparosa.
Con el tren en marcha, John recoge explosivos y sube al vagón blindado. Coloca una carga en la escotilla, se aparta y entra después de que la explosión la reviente. En el interior, abre la caja fuerte y toma lo que contiene. John vuelve con Reyes, quien le dice que pronto conseguirá la información que necesita, y lo deja junto al tren, a las afueras de Casa Madrugada.

Curiosidades
- ◆The name of this mission is inspired by the 1903 silent film The Great Train Robbery.
- ◆The plot of robbing a train loaded with ammunition is perhaps an allusion to Sam Peckinpah's film The Wild Bunch, which Rockstar noted as one of the films that inspired the game. In the film, however, the characters rob a train for a character not very unlike Colonel Allende.
- ◆This is the only time that a train is seen pulling more than five cars.
- ◆It always rains during this mission, possibly because the darkness of the clouds can allow John to approach more surreptitiously, and that the thunderstorm can help hide unnecessary noises.
- ◆This is the only Reyes mission which does not involve killing an antagonist. It is also the only Reyes mission that doesn't reward the player an Achievement/Trophy.
- ◆This is one of the few missions to require the player to neutralize an enemy without alerting anyone to their presence, another being At Home With Dutch.
- ◆If the player shoots while inside the armored wagon Reyes will make a comment about it like "Are you shooting imaginary soldiers?".
- ◆After the train is uncoupled at Chuparosa, it rolls westwards, down the hill. The uncoupled train never actually stops, and continues to gain speed indefinitely. Using mods, it's possible to watch the unmanned train cross into the USA, making a loop around Gaptooth Breach and Tumbleweed, passing through Benedict Point, then re-entering Mexico. It will even pass its point of origin in Chuparosa, clipping through the stationary train, and zooming off through Mexico, back into the USA. By the time it has passed MacFarlane's Ranch and entered the tunnel in the Redemption Mountains, it is too fast to keep up with.
