John Marston llega a Agave Viejo y exige a Abraham Reyes saber cómo piensa capturar a Javier Escuella. Reyes le explica que Escuella está en El Presidio, y que tomar el fuerte del Ejército Mexicano también lo pondrá en manos rebeldes. John sube con Reyes a una carreta cargada de TNT. Cerca del fuerte, Reyes enciende la mecha y espolea a los caballos. John salta antes de que la carreta atraviese las puertas de El Presidio y permita entrar a los rebeldes.
John combate a los soldados mexicanos que salen a defender el fuerte y registra sus edificios en busca de Escuella. Lo encuentra arriba, en los barracones. Escuella promete conducirlo hasta Bill Williamson y Dutch van der Linde a cambio de su vida, pero le arroja una caja encima y huye por una ventana. John lo persigue fuera de El Presidio, le da alcance y lo captura o lo mata.
Cuando Escuella queda fuera de combate, los refuerzos del Ejército Mexicano se acercan a El Presidio. John maneja un cañón y destruye las patrullas y las carretas que avanzan hacia el fuerte. Después recoge a Escuella y lo lleva hasta la costa, donde Reyes bromea sobre el reencuentro de antiguos amigos.
En una isla al norte de Ramita de la Baya, John se reúne con Edgar Ross y Archer Fordham. Entrega a Escuella a los agentes, o su cadáver si lo ha matado. Ross y Fordham le indican que vaya tras Bill Williamson y que viaje a Blackwater, pues creen que Dutch van der Linde se encuentra allí.

Curiosidades
- ◆If the player is replaying the mission, Reyes will always say that John didn't kill De Santa during the previous mission.
- ◆In this mission, John obtains the Bolt Action Rifle if he hasn't bought it before.
- ◆There has been a glitch reported when John Marston lassoes Escuella, just before he hogties him on the road leading out of El Presidio. This has also occurred when you use Dead Eye to shoot Escuella's horse from underneath him. The glitch is strange, as even though Escuella is on the ground lassoed, both Escuella and John Marston continue talking as before when the chase for Escuella was on.
- ◆When the army sends reinforcements, even if the player does nothing, the other rebels are entrenched well enough to handle the few soldiers attacking El Presidio, thus making it near impossible to not obtain victory at this point in the mission.
- ◆This is the only mission in which John cries (if the player kills Escuella).
- ◆Once the player completes this mission and has not started the next mission, there is a glitch in Casa Madrugada. Wild animals will spawn very often and will not attack the player.
- ◆Edgar Ross and Archer Fordham manage to get through New Austin despite the bridge from West Elizabeth to New Austin being unusable at the time of the mission.
- ◆In the Game of the Year Edition, the woman Reyes is having sex with will have her breasts covered up by her clothes, unlike in the original version where they are visible. This censorship was reverted in later versions of the game.
