John Marston chega a Agave Viejo e cobra de Abraham Reyes o plano para capturar Javier Escuella. Reyes diz que Escuella está em El Presidio e que atacar o forte do Exército Mexicano também pode entregá-lo aos rebeldes. John sobe com Reyes numa carroça carregada de TNT. Perto do forte, Reyes acende o pavio e lança os cavalos adiante. John salta antes que a carroça atravesse os portões de El Presidio e abra passagem para os rebeldes.
John enfrenta os soldados mexicanos que defendem o forte e vasculha seus prédios atrás de Escuella. Ele o encontra no andar de cima do alojamento. Escuella promete levar John até Bill Williamson e Dutch van der Linde se for poupado, mas derruba uma caixa sobre ele e foge pela janela. John o persegue para fora de El Presidio, alcança-o e o captura vivo ou o mata.
Depois que Escuella é detido, reforços do Exército Mexicano avançam sobre El Presidio. John assume um canhão e destrói as patrulhas e carroças que se aproximam, impedindo que retomem o forte. Em seguida, ele pega Escuella e o leva até a costa, onde Reyes comenta a reunião de velhos amigos.
Numa ilha ao norte de Ramita de la Baya, John encontra Edgar Ross e Archer Fordham. Ele entrega Escuella aos agentes, ou mostra o corpo caso o tenha matado. Ross e Fordham dizem que o próximo alvo é Bill Williamson e mandam John seguir para Blackwater, onde acreditam que Dutch van der Linde esteja.

Curiosidades
- ◆If the player is replaying the mission, Reyes will always say that John didn't kill De Santa during the previous mission.
- ◆In this mission, John obtains the Bolt Action Rifle if he hasn't bought it before.
- ◆There has been a glitch reported when John Marston lassoes Escuella, just before he hogties him on the road leading out of El Presidio. This has also occurred when you use Dead Eye to shoot Escuella's horse from underneath him. The glitch is strange, as even though Escuella is on the ground lassoed, both Escuella and John Marston continue talking as before when the chase for Escuella was on.
- ◆When the army sends reinforcements, even if the player does nothing, the other rebels are entrenched well enough to handle the few soldiers attacking El Presidio, thus making it near impossible to not obtain victory at this point in the mission.
- ◆This is the only mission in which John cries (if the player kills Escuella).
- ◆Once the player completes this mission and has not started the next mission, there is a glitch in Casa Madrugada. Wild animals will spawn very often and will not attack the player.
- ◆Edgar Ross and Archer Fordham manage to get through New Austin despite the bridge from West Elizabeth to New Austin being unusable at the time of the mission.
- ◆In the Game of the Year Edition, the woman Reyes is having sex with will have her breasts covered up by her clothes, unlike in the original version where they are visible. This censorship was reverted in later versions of the game.
