John Marston encontra Abraham Reyes em Agave Viejo, onde Reyes pede ajuda para tomar um trem de suprimentos do Exército Mexicano. Os dois cavalgam sob a chuva até Chuparosa. Reyes explica que precisam desacoplar os vagões traseiros antes da partida, separando a locomotiva e o vagão blindado que pretendem capturar. Ele provoca uma distração enquanto John entra na plataforma sem alertar os soldados.
John elimina em silêncio os guardas ao longo do trem, chega ao engate e solta os últimos vagões. Enquanto eles descem pelos trilhos, John enfrenta os soldados que protegem o restante da composição, inclusive o homem na Gatling gun. Depois de limpar os vagões à frente, ele solta o freio da locomotiva e assume seu controle. Reyes alcança o trem quando ele já deixa Chuparosa para trás.
Com o trem em movimento, John pega explosivos e sobe no vagão blindado. Ele instala uma carga na escotilha, se afasta e entra quando a explosão a abre. Lá dentro, arromba o cofre e recolhe o que está guardado. John volta para Reyes, que promete conseguir em breve as informações de que ele precisa, e o deixa ao lado do trem, do lado de fora de Casa Madrugada.

Curiosidades
- ◆The name of this mission is inspired by the 1903 silent film The Great Train Robbery.
- ◆The plot of robbing a train loaded with ammunition is perhaps an allusion to Sam Peckinpah's film The Wild Bunch, which Rockstar noted as one of the films that inspired the game. In the film, however, the characters rob a train for a character not very unlike Colonel Allende.
- ◆This is the only time that a train is seen pulling more than five cars.
- ◆It always rains during this mission, possibly because the darkness of the clouds can allow John to approach more surreptitiously, and that the thunderstorm can help hide unnecessary noises.
- ◆This is the only Reyes mission which does not involve killing an antagonist. It is also the only Reyes mission that doesn't reward the player an Achievement/Trophy.
- ◆This is one of the few missions to require the player to neutralize an enemy without alerting anyone to their presence, another being At Home With Dutch.
- ◆If the player shoots while inside the armored wagon Reyes will make a comment about it like "Are you shooting imaginary soldiers?".
- ◆After the train is uncoupled at Chuparosa, it rolls westwards, down the hill. The uncoupled train never actually stops, and continues to gain speed indefinitely. Using mods, it's possible to watch the unmanned train cross into the USA, making a loop around Gaptooth Breach and Tumbleweed, passing through Benedict Point, then re-entering Mexico. It will even pass its point of origin in Chuparosa, clipping through the stationary train, and zooming off through Mexico, back into the USA. By the time it has passed MacFarlane's Ranch and entered the tunnel in the Redemption Mountains, it is too fast to keep up with.
