John Marston arriva ad Agave Viejo e incalza Abraham Reyes sul piano per catturare Javier Escuella. Reyes dice che Escuella si trova a El Presidio e che un assalto al forte dell'Esercito Messicano potrebbe anche consegnarlo ai ribelli. John si unisce a Reyes su un carro carico di TNT. Vicino al forte, Reyes accende la miccia e lancia i cavalli in avanti. John salta giù mentre il carro sfonda i cancelli di El Presidio e apre la strada ai ribelli.
John combatte i soldati messicani che entrano nel forte e perquisisce i suoi edifici in cerca di Escuella. Lo trova al piano superiore della caserma. Escuella si offre di condurre John da Bill Williamson e Dutch van der Linde se gli risparmierà la vita, ma poi gli scaraventa addosso una cassa e fugge da una finestra. John lo insegue oltre El Presidio, lo raggiunge e lo cattura vivo oppure lo uccide.
Dopo che Escuella è stato sistemato, i rinforzi dell'Esercito Messicano si avvicinano a El Presidio. John prende il controllo di un cannone e distrugge le pattuglie e i carri di rifornimenti in arrivo prima che possano riprendere il forte. Poi recupera Escuella e lo porta alla riva, dove Reyes accoglie la cattura con una battuta su vecchi amici che si incontrano di nuovo.
Su un'isola a nord di Ramita de la Baya, John incontra Edgar Ross e Archer Fordham. Consegna Escuella agli agenti, oppure ne presenta il corpo se lo ha ucciso. Ross e Fordham dicono a John che Bill Williamson è il suo prossimo obiettivo, poi gli ordinano di recarsi a Blackwater, dove credono si possa trovare Dutch van der Linde.

Curiosità
- ◆If the player is replaying the mission, Reyes will always say that John didn't kill De Santa during the previous mission.
- ◆In this mission, John obtains the Bolt Action Rifle if he hasn't bought it before.
- ◆There has been a glitch reported when John Marston lassoes Escuella, just before he hogties him on the road leading out of El Presidio. This has also occurred when you use Dead Eye to shoot Escuella's horse from underneath him. The glitch is strange, as even though Escuella is on the ground lassoed, both Escuella and John Marston continue talking as before when the chase for Escuella was on.
- ◆When the army sends reinforcements, even if the player does nothing, the other rebels are entrenched well enough to handle the few soldiers attacking El Presidio, thus making it near impossible to not obtain victory at this point in the mission.
- ◆This is the only mission in which John cries (if the player kills Escuella).
- ◆Once the player completes this mission and has not started the next mission, there is a glitch in Casa Madrugada. Wild animals will spawn very often and will not attack the player.
- ◆Edgar Ross and Archer Fordham manage to get through New Austin despite the bridge from West Elizabeth to New Austin being unusable at the time of the mission.
- ◆In the Game of the Year Edition, the woman Reyes is having sex with will have her breasts covered up by her clothes, unlike in the original version where they are visible. This censorship was reverted in later versions of the game.
