John Marston arrive à Agave Viejo et presse Abraham Reyes de lui révéler le plan pour capturer Javier Escuella. Reyes affirme qu'Escuella se trouve à El Presidio, et qu'un assaut contre le fort de la Mexican Army pourrait également le faire tomber aux mains des rebelles. John rejoint Reyes dans un chariot chargé de TNT. Près du fort, Reyes allume la mèche et lance les chevaux vers l'avant. John saute du chariot tandis que celui-ci défonce les portes de El Presidio et ouvre la voie aux rebelles.
John combat les soldats de la Mexican Army qui pénètrent dans le fort et fouille ses bâtiments à la recherche d'Escuella. Il le trouve à l'étage de la caserne. Escuella propose de conduire John jusqu'à Bill Williamson et Dutch van der Linde si John lui laisse la vie sauve, mais il lui pousse alors une caisse dessus et s'échappe par une fenêtre. John le poursuit au-delà de El Presidio, le rattrape et le capture vivant ou le tue.
Des renforts de la Mexican Army approchent de El Presidio après qu'Escuella a été neutralisé. John prend le contrôle d'un canon et détruit les patrouilles et chariots de ravitaillement qui arrivent avant qu'ils ne puissent reprendre le fort. Il récupère ensuite Escuella et le transporte jusqu'au rivage, où Reyes accueille cette capture par une plaisanterie sur de vieux amis qui se retrouvent.
Sur une île au nord de Ramita de la Baya, John rencontre Edgar Ross et Archer Fordham. Il remet Escuella aux agents, ou leur présente son corps s'il l'a tué. Ross et Fordham annoncent à John que Bill Williamson est sa prochaine cible, puis lui ordonnent de se rendre à Blackwater, où ils pensent pouvoir trouver Dutch van der Linde.

Anecdotes
- ◆If the player is replaying the mission, Reyes will always say that John didn't kill De Santa during the previous mission.
- ◆In this mission, John obtains the Bolt Action Rifle if he hasn't bought it before.
- ◆There has been a glitch reported when John Marston lassoes Escuella, just before he hogties him on the road leading out of El Presidio. This has also occurred when you use Dead Eye to shoot Escuella's horse from underneath him. The glitch is strange, as even though Escuella is on the ground lassoed, both Escuella and John Marston continue talking as before when the chase for Escuella was on.
- ◆When the army sends reinforcements, even if the player does nothing, the other rebels are entrenched well enough to handle the few soldiers attacking El Presidio, thus making it near impossible to not obtain victory at this point in the mission.
- ◆This is the only mission in which John cries (if the player kills Escuella).
- ◆Once the player completes this mission and has not started the next mission, there is a glitch in Casa Madrugada. Wild animals will spawn very often and will not attack the player.
- ◆Edgar Ross and Archer Fordham manage to get through New Austin despite the bridge from West Elizabeth to New Austin being unusable at the time of the mission.
- ◆In the Game of the Year Edition, the woman Reyes is having sex with will have her breasts covered up by her clothes, unlike in the original version where they are visible. This censorship was reverted in later versions of the game.
