John Marston retrouve Landon Ricketts autour d'un verre à Chuparosa et lui demande des informations sur Javier Escuella. Ricketts n'en a aucune, mais lui demande pourquoi John tient tant à le retrouver. John raconte les raisons qui l'ont poussé à quitter son ancien gang. Luisa Fortuna fait alors irruption dans le saloon avec la nouvelle que Colonel Allende a condamné à mort, sans procès, des écrivains et des fonctionnaires innocents.
John et Ricketts quittent Chuparosa pour Escalera afin d'intercepter le convoi de prisonniers. Lorsque les chariots apparaissent sur la route, ils attaquent les soldats qui les escortent. John élimine les gardes et les conducteurs sans détruire les chariots, puis prend le contrôle du véhicule de tête. Ricketts à ses côtés, il dirige le convoi vers la frontière et emmène les prisonniers vers le nord.
Des soldats mexicains bloquent le pont menant à New Austin et attaquent le convoi à coups de feu, de bouteilles incendiaires et avec une Gatling gun. John se fraye un passage à travers l'embuscade, utilisant les explosifs du pont contre les assaillants tandis que les chariots traversent vers Ramita de la Baya. Du côté américain, les prisonniers sont libérés. Ricketts prend en charge la suite de leur voyage et se sépare de John.

Anecdotes
- ◆Landon Ricketts may have never come back from America to Mexico once he escorted the prisoners to the U.S.A. at the end of the mission, since in the newspaper article (published in 1914) about Landon's death, it says that he spent the last three years of his life in the U.S.A., making the year he left Mexico 1911, which is also the year in which he escorts the prisoners into the U.S.A.
- ◆This title is kind of a play on Mexican Train, which is a game played with number dominoes.
- ◆There is a dialogue continuity conflict in this mission. There is a bit where Ricketts admonishes Marston for "jumping fences" and helping both the rebels and Allende. However, this mission can be completed without having done any of the missions given by Vincente de Santa, Luisa Fortuna, or Abraham Reyes.
- ◆At the end of this mission, John and Ricketts part ways, so Ricketts never actually helped John in finding Williamson or Escuella.
- ◆Rob Wiethoff, the voice actor for John Marston, has said that this is his favorite mission in the game.
- ◆The prisoners rescued by Marston are Harris Carruthers, Eudoro de la Barra, Fermin Ichinaga and Porfirio Gutierrez. Even if the player drives the wagon with Ichinaga and Ontiveros, they will still get the cutscene with Carruthers and De la Barra.
- ◆None of the prisoners seem to be the "prominent writer, Castilla" or the "local official" who Luisa identified. However, this is likely just a developer oversight.
- ◆Unluckily for Ichinaga, this is actually the second time that John and Marston have to rescue him, the first being in the mission The Gunslinger's Tragedy.
- ◆At the start of the mission, after the cutscene in which Luisa appears, she can be seen walking into Chuparosa church, where she prays beside the altar.
