La Del Lobo Gang è una banda composta soprattutto da fuorilegge, esuli e opportunisti messicani, anche se tra le sue file cavalcano pure degli americani. Il risentimento lasciato dalla guerra messicano-statunitense alimenta la loro ostilità verso il governo degli Stati Uniti. Predano New Austin da Thieves' Landing, la loro base principale, e mantengono covi a Fort Mercer, Twin Rocks, Gaptooth Breach e Solomon's Folly. Le loro attività comprendono rapine, estorsioni e omicidi, e la debolezza delle forze dell'ordine durante l'epidemia di colera lascia gran parte della regione esposta.
Nel 1898, Marshal Tom Davies manda il protagonista di Online e Lee a Tall Trees sulle tracce di Alfredo Montez. Il gruppo trova e uccide invece Jorge Montez, scambiando il fratello di Alfredo per il proprio bersaglio. Ulteriori scontri con i Del Lobos a Van Horn Trading Post e in un accampamento vicino portano alla cattura di Alfredo presso Butcher Creek. Viene detenuto a Valentine, dove Davies lo giustizia senza processo prima che la gang possa liberarlo. Il protagonista, Davies, Sheriff Curtis Malloy e gli abitanti di Valentine respingono il raid, poi salvano Davies e Sheriff Sam Freeman dai Del Lobos a Tumbleweed.
Carmela "La Muñeca" Montez assume il comando dopo la morte di Alfredo. Si nasconde con i suoi seguaci a Hanging Dog Ranch, mentre Barbarella Alcazar si accampa con membri della gang a Gaptooth Breach. Entrambe diventano bersagli di taglie per il protagonista di Online. Nel 1899, Arthur Morgan, o John Marston, incontra Flaco Hernández e i suoi seguaci Del Lobo a Cairn Lodge, in Ambarino, mentre è alla ricerca di pistoleri. Dopo la sparatoria, Flaco affronta Arthur o John in duello e muore.
Nel 1907, Sadie Adler e John catturano Ramón Cortez e lo portano allo Scarlett Meadows Sheriff's Office di Rhodes. I Del Lobos lo liberano, ma Sadie e John inseguono la gang, uccidono i soccorritori e riprendono Ramón in custodia. John può in seguito consegnare o uccidere il fratello di Ramón e capo della gang, Esteban Cortez, e può catturare Joaquín Arroyo. - ◆The Del Lobo Gang in several respects resembles the real-life Five Joaquins Gang. Similarly, it was perceived to have been led by predominantly Mexico-born men who were family members.

- ◆A newspaper article mentions how Del Lobos terrorize Valentine and Strawberry. While they can be seen assaulting Valentine in Red Dead Online during the story mission "Bring a Goddamn Posse", members of the gang can not be encountered near Strawberry.
- ◆The Del Lobos are the most widespread gang featured in Red Dead Redemption 2 and Online, as they appear to have a presence in all five states. They are most common in New Austin, but also appear in Ambarino (in "The Noblest of Men, and a Woman"), Lemoyne (in "A Quick Favor for an Old Friend"), West Elizabeth (in "Where Your Morals Lead You"), and New Hanover (in "Highly Illegal And Highly Moral" and "Bring a Goddamn Posse").
- ◆Following Update 1.15, the Del Lobos became the gang with the highest number of hideouts (4 in total; 5 with the Ultimate/Special Edition).
- ◆A random conversation can be heard between Del Lobo members where some of them are thinking of heading back to Mexico and possibly working for the army, while some would opt to stay in the US.
- ◆The group of Del Lobos that the player encounters east and south of Armadillo have custom handguns and rifles with gold plated parts, ebony grips and expensive varnishes.
- ◆The Del Lobos are the only group of NPCs in Red Dead Redemption 2 known to throw dynamite and fire bottles at the player. These weapons can also be looted from their corpses.
- ◆After clearing out Thieves Landing a couple of times, and returning when the Del Lobos present, the player can eavesdrop on the members grieving for the loss of their comrades, putting the blame on an unseen group of New Austin vigilantes and an unnamed cowboy.
