The Del Lobo Gang is a band of mostly Mexican outlaws, exiles and opportunists, though Americans also ride with them. Resentment left by the Mexican-American War feeds their hostility toward the United States government. They prey on New Austin from Thieves' Landing, their principal base, and hold hideouts at Fort Mercer, Twin Rocks, Gaptooth Breach and Solomon's Folly. Their work includes robbery, extortion and murder, and the weak law enforcement during the cholera outbreak leaves much of the region exposed.
In 1898, Marshal Tom Davies sends the Online protagonist and Lee into Tall Trees after Alfredo Montez. The party finds and kills Jorge Montez instead, mistaking Alfredo's brother for their target. Further clashes with Del Lobos in Van Horn Trading Post and at a nearby camp lead to Alfredo's capture near Butcher Creek. He is held in Valentine, where Davies executes him without trial before the gang can free him. The protagonist, Davies, Sheriff Curtis Malloy and Valentine residents repel the raid, then rescue Davies and Sheriff Sam Freeman from Del Lobos at Tumbleweed.
Carmela "La Muñeca" Montez takes charge after Alfredo's death. She hides with her followers at Hanging Dog Ranch, while Barbarella Alcazar camps with gang members at Gaptooth Breach. Both become bounty targets for the Online protagonist. In 1899, Arthur Morgan, or John Marston, encounters Flaco Hernández and his Del Lobo followers at Cairn Lodge in Ambarino while seeking gunslingers. After the shootout, Flaco faces Arthur or John in a duel and dies.
By 1907, Sadie Adler and John capture Ramón Cortez and bring him to the Scarlett Meadows Sheriff's Office in Rhodes. Del Lobos break him free, but Sadie and John pursue the gang, kill the rescuers and take Ramón back into custody. John can later bring in or kill Ramón's brother and gang leader, Esteban Cortez, and can capture Joaquín Arroyo. In 1911, Del Lobos no longer occupy their old territory in the southern United States or northern Mexico.

- ◆In Red Dead Online, the online protagonist can interact with the Del Lobo NPCs in free roam as long as the player does not attack them.
- ◆The Del Lobo Gang in several respects resembles the real-life Five Joaquins Gang. Similarly, it was perceived to have been led by predominantly Mexico-born men who were family members.
- ◆Del Lobo means 'of the wolf' in Spanish.
- ◆They share the name with Del Lobo Rock, a natural land formation.
- ◆A newspaper article mentions how Del Lobos terrorize Valentine and Strawberry. While they can be seen assaulting Valentine in Red Dead Online during the story mission "Bring a Goddamn Posse", members of the gang can not be encountered near Strawberry.

They are most common in New Austin, but also appear in Ambarino (in "The Noblest of Men, and a Woman"), Lemoyne (in "A Quick Favor for an Old Friend"), West Elizabeth (in "Where Your Morals Lead You"), and New Hanover (in "Highly Illegal And Highly Moral" and "Bring a Goddamn Posse").
- ◆Following Update 1.15, the Del Lobos became the gang with the highest number of hideouts (4 in total; 5 with the Ultimate/Special Edition).
- ◆A random conversation can be heard between Del Lobo members where some of them are thinking of heading back to Mexico and possibly working for the army, while some would opt to stay in the US.
- ◆The group of Del Lobos that the player encounters east and south of Armadillo have custom handguns and rifles with gold plated parts, ebony grips and expensive varnishes.
- ◆The Del Lobos are the only group of NPCs in Red Dead Redemption 2 known to throw dynamite and fire bottles at the player. These weapons can also be looted from their corpses.
- ◆After clearing out Thieves Landing a couple of times, and returning when the Del Lobos present, the player can eavesdrop on the members grieving for the loss of their comrades, putting the blame on an unseen group of New Austin vigilantes and an unnamed cowboy.
