Der Vapid Peyote ist ein zweitüriges, zweisitziges Sports Classics Coupé, dessen Design von amerikanischen Autos der 1950er Jahre inspiriert ist. In Grand Theft Auto V behält er das Modell aus Grand Theft Auto IV mit umfangreichen Chromleisten, übereinander angeordneten runden Scheinwerfern und Rückleuchten, einem breiten Kühlergrill, Heckflossen und Rädern im Drahtspeichenstil. Sein Dach kann eine zweite Farbe tragen, während ein Continental-Kit als Los Santos Customs-Modifikation erhältlich ist. Einige Exemplare verfügen zudem über Zebrapolster und schwarze Plüschwürfel.

Der Peyote steht nahe Michaels Anwesen und im Süden von East Los Santos geparkt und kann im Verkehr in Vespucci auftauchen. Während Random Event - Car Theft 1 erscheint ein auffälliges rosa Exemplar. Das Fahrzeug kann behalten werden, nachdem es dem Dieb abgenommen wurde, statt es seinem Besitzer zurückzugeben. Modifizierte dachlose Peyotes erscheinen auch rund um El Burro Heights, besonders auf dem Bus-Schrottplatz an der South Shambles Street und Hanger Way, meist nachts.

Der Peyote ist schwerer als sein früheres Gegenstück, was seine Beschleunigung verringert und seine Höchstgeschwindigkeit senkt, obwohl sein Motor derselbe bleibt. Sein Bremsweg ist kürzer. In Los Santos wird er von den Los Santos Vagos, Families und Ballas auch als Lowrider genutzt. Ihre dachlosen Varianten besitzen entchromte Stoßfänger, Ersatzräder, Lead-Sled-Kühlergrills, Cowl-Induction-Motorhauben, Lowrider-Räder, Rennsportfederungen und gangspezifische Gestaltungen. Franklin kann einen Families Peyote nehmen, ohne angegriffen zu werden.

◆"Peyote" is the real-life common name of a North American breed of cactus, famous for its powerful psychoactive properties. A badge emblazoned with an image of the peyote cactus is placed on side of the hardtop variant's rear pillars.
◆Despite having two textures of its "V10 supercharged" badge for each side, the text is mistakenly mirrored on one side. This was fixed in GTA V.
◆A Rockstar Games emblem can be seen on Peyotes with zebra seats.
◆One of the modifications of the Peyote includes a pair of fuzzy dice on the mirror, just like the Yardie Lobo from GTA III and the Bickle'76 from GTA Liberty City Stories. This is the third car in the series to feature a pair of fuzzy dice on the mirror.
◆However, the pair of fuzzy dice was removed in the enhanced version.
◆The "Los Santos Plaque" is not spelled the same way as the options menu in Los Santos Customs. It actually reads "Low Santos", referencing its low rider qualities.
◆The gang and non-gang modified topless Peyotes can rarely have side windows. This also applies to the gang and non-gang modified topless Mananas.
◆The description on Benny's Original Motor Works refers to how the Peyote was originally only obtainable by stealing one on the streets.
◆The Route 68 poster seems very similar to the GTA Online Social Club preview, only with different backgrounds; the Route 68 being on said road, while the Social Club preview is seen on the city streets.
