John Marston reaches Luisa Fortuna's camp at Campo Mirada, where she reports that Captain Vincente de Santa is directing a massacre at Sepulcro. Luisa asks John to stop him and his soldiers. John rides with the rebels toward the cemetery, ordering them to hold their fire until he gives the word. Gunfire from Sepulcro interrupts the approach, and John hurries ahead as De Santa's men kill the rebels inside.
John enters Sepulcro and fights through the Mexican soldiers, keeping De Santa alive. When the captain flees, John pursues him across the cemetery, catches him with a lasso and brings him back to the rebels. He beats De Santa for the whereabouts of Bill Williamson and Javier Escuella. De Santa says Javier is at Casa Madrugada. John can kill De Santa himself or leave him to Reyes' rebels.
John rides to Casa Madrugada and finds its streets apparently empty. As he searches the settlement for Javier, Mexican soldiers emerge from the upper floors and ground floor of the large building ahead. John kills them, then speaks with a prostitute. She tells him Javier has not been in Casa Madrugada for months, and that De Santa has sent him into a trap.

Trivia
- โDuring the mission "The Gates of El Presidio," Abraham Reyes will talk to you about the death of Captain De Santa. His quotes will change depending on whether the player or the rebels killed De Santa.
- โThe shootout sequence in this mission is very similar to another Rockstar Games title mission, "Los Sepulcros" from Grand Theft Auto: San Andreas, and even shares a variation of its name with the cemetery.
- โWhen De Santa is running from Marston he can lead him the same way as cattle if heโs on his horse.
- โIf the rebels and De Santa are in the right spot, when De Santa pulls out his weapon he will engage with the rebels and ignore Marston.
- โDe Santa has infinite revolvers and rifles when he start shooting at John. No matter how many time you disarm him he will pull out another weapon.
