Sean MacGuire grandit dans le comté de Donegal, en Irlande, fils du hors-la-loi Fenian Darragh MacGuire. Après que Darragh s'est enfui avec lui en Amérique avant de mourir plus tard, Sean est envoyé dans une maison de correction, ce qui ne le change guère. Il rencontre Dutch van der Linde et Hosea Matthews dans un bar de North Elizabeth, les suit dans une ruelle pour voler la montre de poche de Dutch, et découvre qu'ils ont vidé son arme. Amusés par sa tentative, ils le nourrissent et l'intègrent au gang Van der Linde.
Dans Red Dead Online, Sean est séparé de ses compagnons et rencontre le protagoniste dans une cabane à Tanner's Reach. Il le tient d'abord en joue, puis se calme et lui transmet des informations sur une diligence de passage. Sean propose ensuite des travaux criminels depuis Tanner's Reach ou un camp près de Quaker's Cove. Durant Blood Money, il travaille au sein du réseau criminel de Guido Martelli et fournit d'autres missions au protagoniste.
Après le braquage du ferry de Blackwater, Sean est assommé, séparé du gang et capturé par des chasseurs de primes menés par Ike Skelding. Josiah Trelawny apprend qu'il est transféré vers une prison fédérale, alors Arthur Morgan, Javier Escuella, Charles Smith et Trelawny partent à sa recherche. Ils trouvent Sean suspendu la tête en bas dans un camp de chasseurs de primes près de Blackwater, le libèrent après une fusillade et rentrent avec lui à Horseshoe Overlook. Ce soir-là, Sean boit avec le gang et part avec Karen Jones. Plus tard, il rejoint Arthur, Charles et John Marston pour le braquage d'un train, pille le wagon à bagages et s'échappe après qu'Arthur l'a sauvé des gardes.
À Clemens Point, Sean accompagne Arthur et Hosea Matthews dans le travail de Catherine Braithwaite contre la famille Gray. Il aide Arthur à infiltrer Caliga Hall, incendie l'exploitation de tabac des Gray et s'enfuit avec la paie prise dans un chariot. Il emmène aussi Arthur pour un braquage de propriété et rejoint Arthur et Mary-Beth Gaskill pour le braquage d'une diligence. À Rhodes, Sean marche avec Arthur, Bill Williamson et Micah Bell vers un prétendu travail pour les Gray. Un tireur embusqué des Gray lui tire dans la tête avant que les autres puissent réagir. Arthur tue Sheriff Gray après l'embuscade et dit à Bill de veiller à ce que Sean reçoive un enterrement digne de ce nom.

- ◆Sean is the first member of the Van der Linde Gang to canonically have an on-screen death (not counting Davey Callander who dies at the time of his introduction).
- ◆After completing "A Short Walk in a Pretty Town", Sean's grave can be found slightly north of Clemens Point.
- ◆His horse, Ennis, shares a name with the county town of County Clare.
- ◆Like most members of the Van der Linde gang, Sean has a custom sidearm. His weapon is a Cattleman Revolver in blued steel with a nickel frame.
- ◆He says the word "fuck" more than any other member of the gang.
- ◆Sean's knife has a date engraved on the handles, 1848, the same year in which a series of revolutions occurred in Europe, and groups of young Irishmen had participation in those revolutions but failed like the great majority in that year. His knife was probably a gift from his family or father.
- ◆Despite being Irish, Sean's surname is spelled the Scottish way, with the prefix "Mac". However, it is not unheard of for Irish names to use the "Mac" prefix as well.
- ◆During a random camp conversation with Kieran, Sean will call him out for mispronouncing Colm O'Driscoll's first name, which is an Irish name. Sean eventually threatens Kieran until he pronounces the name correctly, despite Kieran insisting that Colm himself uses the American pronunciation.
- ◆Sean is one of three members to not have a winter outfit. The other two are Cleet and Josiah Trelawny.
- ◆Sean has an unused outfit consisting of a black leather stitched up duster coat, similar to the one worn by members of the gang during robberies.
- ◆When MacGuire sustains a head injury during "Pouring Forth Oil IV", Arthur says to him, "least you ain't got a hole in it", foreshadowing how Sean would later die.
- ◆If Arthur antagonizes the gang members enough, Sean is one of the characters who may punch him out of the camp. The others are Lenny, Javier, Bill, John, and Charles.
- ◆Sean, Sadie Adler, Joe, and Trelawny are the only members of the Van der Linde gang to appear in Red Dead Online.
- ◆Beside Tanner's Reach, Sean can also be found at camp southwest of Blackwater in Red Dead Online.
- ◆During his first encounter in Red Dead Online, the animation shows Sean cocking the hammer of his revolver but the hammer does not move. This is due to an oversight where the gun was not properly animated for the scene.
- ◆While having sex with Karen after being rescued from Blackwater, Sean mentions that one of his sexual acts (which annoys Karen) was "an old trick me Da taught me". This joke shows how Sean views his father's identity as a key part of his own life.
- ◆During Pouring Forth Oil IV, Sean reloads his Cattleman Revolver incorrectly during the first cutscene while he is practicing his gunslinging.
- ◆The Cattleman Revolver is reloaded via half-cocking the hammer and loading bullets through the loading gate. Sean's revolver swung out to the side, similar to the Double Action Revolver.
- ◆This is the only time Sean is seen reloading his revolver this way. Before and after the cutscene, Sean will start to reload correctly again.
