O Lobo representa Canis lupus em Red Dead Redemption 2 e aparece nas variantes Lobo-cinzento, Lobo-da-madeira e Lobo Lendário. É um canídeo grande e esguio, de focinho largo, orelhas curtas, tronco curto e cauda longa. Em comparação com coiotes e raposas, é maior e mais pesado, com pernas mais longas que permitem avançar depressa pela neve profunda.
Lobos vivem, viajam e caçam em alcateias, geralmente de três a seis animais. Caçam alces, alces-americanos, cervos, cabras-montesas, castores e presas menores, além de se alimentarem de carcaças. O uivo de uma alcateia pode anunciar sua investida, enquanto os sobreviventes fogem ganindo depois de perder a maior parte do grupo. São mais ativos ao amanhecer e ao entardecer, e aparecem em Aurora Basin e Hanging Dog Ranch.
Um lobo morto fornece Wolf Fur, Wolf Heart, Big Game Meat e Wolf Carcass. A carne é usada como alimento, e as peles e os corações entram em trabalhos de criação. O Trapper usa uma pele de lobo perfeita para produzir o Wilderness Vest, o Wolf Coat e o Legendary Moose Hunting Jacket.
